dc.description.abstractThe medium components were optimized by using a statistical experimental design approach for increasing chitinase production from the Antarctic bacterium Sanguibacter antarcticus KOPRI 21702. To increase the production of psychrophilic chitinase, significant carbon and nitrogen sources were determined and other elements were chosen by using a two-level Plackett-Burman statistical design technique. In the first optimization step, glycerol and peptone were selected as main carbon and nitrogen sources, respectively. In the second step, the influence of fifteen components included in Marine Broth medium on chitinase production was evaluated. As a result, NaCl, Fe(C6H5O7), MgCl2, Na2SO4, H3BO3, and CaCl2 were found to have a positive influence on the chitinase production, while KCl, KBr, and NH4NO3 had a negative influence. In the third step, the concentrations of NaCl, Fe(C6H5O7) and MgCl2 were further optimized. The maximum chitinase activity obtained was 89 U/L using 10 g/L glycerol, 5 g/L peptone, 23 g/L NaCl, 0.01 g/L Fe(C6H5O7), and 2.5 g/L MgCl2.
